BPC-157
Pentadecapeptide (15 amino acid sequence) derived from a protein found in human gastric juice. Also known as PL-10 or Pentadecapeptide BPC 157. Does not bind a single high-affinity receptor — instead modulates the nitric-oxide system, VEGF expression, and growth-factor signaling involved in tissue repair.
TB-500 (Thymosin β4 fragment)
Synthetic 23-amino-acid fragment of Thymosin β4, a naturally occurring peptide involved in actin sequestration and tissue repair. TB-500 promotes cell migration to injury sites, supports angiogenesis, and reduces inflammatory scarring during the repair process.
GHK-Cu (Copper tripeptide-1)
Glycyl-L-histidyl-L-lysine bound to copper(II). Naturally occurring tripeptide found in human plasma, saliva, and urine. Blood levels decline significantly with age — from ~200 ng/mL at age 20 to ~80 ng/mL by age 60 — correlating with reduced tissue repair capacity.
Acetate buffer
Maintains reconstituted solution at pH 5.0–5.5 — the range in which the peptide is most chemically stable. Degrades to water and acetate; renally cleared.
Mannitol
USP-grade bulking agent used during lyophilization. Produces a uniform cake, prevents peptide collapse during reconstitution, and has no pharmacologic activity at the doses used.
Nitric oxide system
Upregulates eNOS activity and counteracts L-NAME-induced vascular dysfunction in animal models.
Collagen & elastin synthesis
Upregulates type I and type III collagen production by dermal fibroblasts. Concurrently increases elastin and glycosaminoglycan synthesis, supporting extracellular matrix restoration.
VEGF / angiogenesis
Increases VEGFR2 expression, associated with accelerated capillary ingrowth at injury sites. Promotes formation of new blood vessels during tissue repair.
Copper-dependent enzymes
Delivers bioavailable copper to lysyl oxidase (collagen crosslinking), superoxide dismutase (antioxidant defense), and cytochrome c oxidase (mitochondrial respiration). Addresses age-related decline in copper-dependent enzyme activity.
Growth factor modulation
Elevates local expression of TGF-β and EGR-1, both implicated in fibroblast recruitment. Enhances collagen organization in healing tendon and ligament tissue.
Anti-inflammatory modulation
Suppresses NF-κB signaling and reduces pro-inflammatory cytokine release (TNF-α, IL-6). Shifts macrophage polarization toward M2 repair phenotype in wound models.
GI mucosa integrity
Attenuates ulcer formation from NSAID, alcohol, and stress models; exact mechanism multifactorial. Supports gut barrier function and may reduce intestinal permeability.
Hair follicle stimulation
Activates dermal papilla cells and upregulates VEGF and noggin expression, extending the anagen phase. Limited human data on oral or systemic dosing; topical effects well-documented.
Serotonergic / dopaminergic
Modulates 5-HT and dopamine turnover in limited animal studies; clinical relevance unclear. May contribute to reported mood and sleep effects, though evidence is preliminary.
Gene expression reset
Microarray studies show GHK-Cu modulates over 4,000 human genes toward a younger expression profile, including DNA repair genes and tumor suppressors. Long-term in vivo relevance still being characterized.
TB-500 · Actin remodeling
Sequesters G-actin and supports actin polymerization at sites of cellular stress, enabling cell migration into damaged tissue. Promotes endothelial cell migration and angiogenesis, complementing the VEGF-driven repair seen with BPC-157.
TB-500 · Anti-fibrotic action
Reduces scarring during repair by modulating TGF-β signaling and matrix metalloproteinase activity. Combined with BPC-157's repair signaling, the pair is associated with cleaner connective tissue regeneration in animal models.
